Abnormal Function of the Bone Marrow Microenvironment in Chronic Myelogenous Leukemia: Role of Malignant Stromal Macrophages

چکیده

The bone marrow microenvironment supports and regulates the proliferation and differentiation of hematopoietic cells. Dysregulated hematopoiesis in chronic myelogenous leukemia (CML) is caused, at least in part, by abnormalities in CML hematopoietic progenitors leading to altered interactions with the marrow microenvironment. The role of the microenvironment itself in CML has not been well characterized. We examined the capacity of CML stroma to support the growth of long-term culture-initiating cells (LTC-IC) obtained from normal and CML marrow. The growth of normal LTC-IC on CML stroma was significantly reduced compared with normal stroma. This did not appear to be related to abnormal production of soluble factors by CML stroma because normal LTC-IC grew equally well in Transwells above CML stroma as in Transwells above normal stroma. In addition, CML and normal stromal supernatants contained similar quantities of both growth-stimulatory (granulocyte colony-stimulating factor (CSF), interleukin-6, stem cell factor, granulocyte-macrophage CSF, and interleukin-1/31 and growth-inhibitory cytokines (transforming growth factor-p, macrophage inflammatory protein-la, and tumor necrosis factor-a). The relative proportion of different cell types in CML and normal stroma was similar. However, polymerase

Advances in Hematopoietic Stem Cell Mobilization and Peripheral Blood Stem Cell Transplantation

Hematopoietic stem/progenitor cells (HSPCs) which give rise to different blood cell types are present within the bone marrow microenvironment, especially in flat bones such as skull, vertebrae, pelvis and chest. Interacting factors such as stromal derived factor-1/CXCR4, very late antigen-4/vascular cell adhesion molecule-1, Lymphocyte function-associated antigen-1/ intercellular adhesion molec...

"Recovery" of In-111 Chloride uptake in spleen and bone marrow, after chemotherapy for acute leukemia

A man with acute myelogenous leukemia, treated with chemotherapy, had a decrease in circulating formed blood elements. Bone marrow biopsy revealed necrosis. Imaging with In-Ill chloride demonstrated activity in the liver, but not in the spleen or bone marrow. Eighteen days later, the circulating blood count had risen; a repeat bone marrow biopsy showed the return of blood cell precursors....

Pregnancy Outcome of Two Patients with Chronic Myelogenous Leukemia Treated with Imatinib

Although chronic myelogenous leukemia in pregnancy is rare, its management and treatment is more difficult and complicated.Treatment of patients with chronic myelogenous leukemia includes bone marrow transplantation, however in less than 30% of patients the donor’s organ would be accepted. To this end, cytotoxic therapy is considered as an alternative therapeutic option. This option provides sa...

Multiple Myeloma Bone Marrow Mesenchymal Stromal Cells Inhibit CD8+ T Cell Function in a Process that May Implicate Fibroblast Activation Protein α

Background: Multiple myeloma (MM) is a malignant plasma cell proliferative disorder with limited immunotherapy treatment because of T cell dysfunction. Objective: To investigate the immunomodulatory function of bone marrow mesenchymal stromal cells (MM-BMSCs) on CD8+ T cells. Methods: Proliferation and cytotoxicity were detected by c...
